No, ReadyBoost relies on the flash properties of the memory card to speed up your PC by using it as a cache for pre-loaded files and such. Loading these onto an external USB hard drive would give similar or worse performance then loading them directly from the internal hard drive, and as such would be utterly pointless.
Also, as a side note, ReadyBoost only works on sticks with at least 512mb of memory, so older sticks with smaller capacities are no use.
